| Background | SIGLEC8 (Sialic acid-binding immunoglobulin-like lectin 8) is an inhibitory cell-surface receptor belonging to the CD33-related Siglec family. It is predominantly expressed on human eosinophils, mast cells, and basophils, where it recognizes specific sialylated glycans and regulates immune-cell activation. The cytoplasmic region of SIGLEC8 contains immunoreceptor tyrosine-based inhibitory motifs that recruit signaling molecules and suppress downstream cellular responses. Engagement of SIGLEC8 can induce apoptosis in eosinophils and inhibit mast-cell activation, making it an important regulator of allergic inflammation and eosinophil-associated diseases. SIGLEC8 has therefore emerged as a promising therapeutic target for conditions such as asthma, chronic rhinosinusitis with nasal polyps, eosinophilic disorders, and mast-cell-mediated diseases. Recombinant SIGLEC8 protein is suitable for glycan-binding studies, receptor–ligand interaction assays, antibody screening, drug discovery, and research on allergic and eosinophilic inflammation. |
