| Background | HRH1 (Histamine receptor H1) is a G-protein coupled receptor (GPCR) that primarily couples to Gq/11 proteins, activating phospholipase C (PLC), increasing IP₃/DAG levels and intracellular Ca²⁺ mobilization. HRH1 is expressed in smooth muscle, endothelial cells, brain, and immune cells, mediating allergic responses, vasodilation, bronchoconstriction, and neurotransmission. Dysregulation of HRH1 signaling is associated with allergies, asthma, inflammation, and CNS disorders, making it a therapeutic target for antihistamines and allergy treatments. |
