| Background | NTSR2 encodes the neurotensin receptor type 2, a G-protein coupled receptor (GPCR) that binds the neuropeptide neurotensin (NTS). It primarily couples to Gq/11 and Gi/o proteins, triggering downstream phospholipase C activation, Ca²⁺ mobilization, and MAPK signaling. NTSR2 is expressed in the central nervous system (notably in the midbrain, hypothalamus, and spinal cord) as well as in peripheral tissues. It mediates neurotensin-induced analgesia, hypothermia, and regulation of dopamine transmission, and contributes to neuronal excitability, pain modulation, and neuroendocrine secretion. Altered NTSR2 expression has been linked to psychiatric disorders, neuroinflammation, and certain cancers, making it a potential therapeutic target in neurobiology and oncology. |
