| Background | HTR2A encodes the 5-hydroxytryptamine (serotonin) receptor 2A, a G-protein coupled receptor (GPCR) that mediates many of serotonin’s effects in the central nervous system and peripheral tissues. It primarily couples to Gq/11 proteins, activating the phospholipase C (PLC)–IP₃–Ca²⁺ signaling pathway, leading to neuronal excitation and modulation of neurotransmitter release. HTR2A is highly expressed in the cerebral cortex, hippocampus, and platelets, and regulates mood, perception, sleep, and cognition. Dysregulation or polymorphisms of HTR2A are associated with psychiatric disorders such as schizophrenia, depression, and anxiety, and it is a pharmacological target of antipsychotics, antidepressants, and hallucinogens (e.g., LSD, psilocybin). |
