| Background | C3AR1 encodes the C3a receptor (C3aR1), a G-protein coupled receptor (GPCR) that binds the complement component C3a, a key anaphylatoxin in the immune system. C3AR1 couples primarily to Gi proteins, leading to inhibition of adenylyl cyclase, Ca²⁺ mobilization, and activation of MAPK and NF-κB signaling. It is expressed on immune cells including neutrophils, monocytes, macrophages, and mast cells, where it mediates chemotaxis, degranulation, cytokine release, and inflammatory responses. C3AR1 plays a critical role in innate immunity, inflammation, and host defense, and dysregulation is implicated in allergic diseases, sepsis, autoimmune disorders, and cancer. |
