| Background | DRD4 (Dopamine receptor D4) is a G-protein coupled receptor (GPCR) that primarily couples to Gi/o proteins, inhibiting adenylyl cyclase and reducing intracellular cAMP. It is mainly expressed in prefrontal cortex, limbic system, and retina, where it modulates cognition, attention, emotion, and behavioral flexibility. Polymorphisms in DRD4 are associated with attention-deficit hyperactivity disorder (ADHD), novelty-seeking behavior, and susceptibility to psychiatric disorders, making it a relevant therapeutic and research target in neuroscience. |
